Angiogenesis
Introduction: Angiogenesis has been shown involved in 26 diseases:
tumor, cardivascular diseases, diabeteic retinopathy, age-related
macular degeneration.
History
- 1962 EGF was dicoverd by Stanley Cohen in opening eye test.
- 1971 PDGF was discovered
- 1971: Surgeon Judah Folkman hypothesizes that tumor growth is
dependent upon angiogenesis.
- 1974 FGF was found by Gospodarowicz
et al in UCSF. Indeed, this was a misnomer. FGF stimulates endothelial
cells more than fibroblasts.
- 1989 VEGF was purified by three groups together: Ferrara's group
in Genentech, Folkman's group in Harvard, Dvorask's group in Harvard.
- 1994 O'Reilly et al found angiostatin as angiognenesis inhibitor
- 1997 O'Reilly et al found endostatin as angiogenesis inhibitor
- 1996,1997 Ang1, Ang2 were cloned by Yancoupolous et al.
- 2004: Bevacizumab (Avastin) was approved by FDA for the
treatment of advanced colorectal cancer.
